Objective:To determine if Diflucan in a clinical trial, compared to an oral formulation of fluconazole, could provide evidence of safety for patients with a yeast infection. Methods: A phase 1b, open-label, multicenter clinical trial, which compared the two topical formulations of fluconazole (Fluconazole and Diflucan) with the oral formulation of fluconazole, for the treatment of yeast infections. Subjects were enrolled in the trial and assigned to either the study treatment or to one of the two treatments. Clinical studies were also performed to assess the safety profile of the two treatments for the treatment of acute and chronic Candida infections. Results: One hundred seventy-one subjects were enrolled, with a mean age of 60.7 years. The mean duration of follow-up was 4.1 months (range 2 to 15). Response was defined as a reduction in the number of yeast-positive cultures in the first day of treatment or, when the infection was confirmed by cultures, a reduction in the number of yeast-positive cultures in the second day of treatment, or, if a change in the number of yeast-positive cultures was required, a decrease in the number of yeast-positive cultures in the third day of treatment. The incidence of treatment-related adverse events was similar between the two treatment groups. Conclusion: Fluconazole is a topical formulation of the most active anti-yeast antifungal agent used in clinical practice, which has a positive effect on the treatment of patients with a yeast infection. Patients with a positive clinical trial result, which could be used to guide future clinical trials, could provide evidence of safety.
